Summary
Green pepper, a nonclimacteric fruit, was selected as an experimental subject to investigate whether low temperature and ethylene are involved in the regulation of mitochondrial respiration (state 3) and alternative oxidase (AOX). Low temperature(s) (12 or 0degreesC) had a stronger stimulatory effect on AOX and mitochondrial respiration compared with ethylene. The effect on AOX was due to the induction of new transcription and translation of AOX and was greater at 0degreesC than at 12degreesC. Exogenous ethylene (100 micronlitres/litre) stimulated mitochondrial respiration, transcription and translation of existing AOX, but did not induce a new isoenzyme of AOX. 1-MCP [methylcyclopropene], an ethylene perception inhibitor, had only a small effect on AOX. This can be explained by the fact that AOX, in a nonclimacteric fruit, may not be regulated by its endogenous ethylene that is always at a very low concentration.
